Acclimatization of 'VR043-43' (Vitis vinifera x Vitis rotundifolia) grapevine rootstock Scientia Agricola

The pre-acclimatization stage can be used to improve micropropagation protocols and increase the yield of produced plants. The influence of sucrose and photon flux density (PFD) levels on the acclimatization of in vitro-grown 'VR043-43' (Vitis vinifera x Vitis rotundifolia) grapevine rootstocks was evaluated. Rooted shoots were obtained from 4-week-old in vitro shoots cultivated in QL (Quoirin and Lepoivre, 1977) culture medium supplemented with 15, 30 and 45 g L-1 of sucrose. RESUMO O objetivo deste trabalho foi analisar as leituras de composição do leite cru por meio de espectrofotometria FTIR, utilizando-se curva de regressão PLS, bem como as contagens de células somáticas e bacteriana total por citometria de fluxo, após adição de amido e sacarose. O leite cru foi adulterado com três concentrações de amido e sacarose (0,1%, 0,5% e 1%), colocado em frascos contendo bronopol ou azidiol, os quais foram armazenados em duas temperaturas (7±2°C e 25±2°C ). Assimilate partitioning in leaves of the raffinose-storing herb Lamium album L.: photosynthesis and carbon partitioning throughout the photoperiod Rev. Bras. Bot.

Lamium album accumulates starch, sucrose and raffinose-family oligosaccharides (RFO) as the major products of photosynthesis. These products were measured in leaves throughout a sixteen-hour photoperiod and under various irradiance conditions. There was continuous accumulation of sucrose and starch. The rate of gas exchange was higher at 500 µEm² s-1 and 900 µEm²s-1 than at 300 µEm² s-1. The rate of photosynthesis did not decline over the sixteen-hour photoperiod, which suggested that there was no short-term feed back inhibition due to sucrose accumulation in this plant. In Lamium album, sucrose and raffinose-family oligosaccharides are the major products of photosynthesis that are stored in leaves. Using gas analysis and 14CO2 feeding, we compared photosynthesis and the partitioning of recently-fixed carbon in plants where sink activity was lowered by excision of flowers and chilling of roots with those where sink activity was not modified. Reduction in sink activity led to a reduction in the maximum rate of photosynthesis, to retention of fixed carbon in source leaves and to the progressive accumulation of raffinose-family oligosaccharides. The filamentous fungus A. phoenicis produced high levels of ?-D-fructofuranosidase (FFase) when grown for 72 hrs under Solid-State Fermentation (SSF), using soy bran moistened with tap water (1:0.5 w/v) as substrate/carbon source. Two isoforms (I and II) were obtained, and FFase II was purified 18-fold to apparent homogeneity with 14% recovery. The native molecular mass of the glycoprotein (12% of carbohydrate content) was 158.5 kDa with two subunits of 85 kDa estimated by SDS-PAGE. Optima of temperature and pH were 55�C and 4.5. Changes in sodium appetite evoked by lesions of the commissural nucleus of the tractus solitarius BJMBR

Ablation of the area postrema/caudal nucleus of the tractus solitarius (NTS) complex increases sodium intake, but the effect of selective lesions of the caudal NTS is not known. We measured depletion-induced sodium intake in rats with electrolytic lesions of the commissural NTS that spared the area postrema. One day after the lesion, rats were depleted of sodium with furosemide (10 mg/kg body weight, sc) and then had access to water and a sodium-deficient diet for 24 h when 1.8% NaCl was offered. Water and saline intakes were measured for 2 h. Saline intake was higher in lesioned than in sham-lesioned rats (mean ± SEM: 20 ± 2 vs 11 ± 3 mL/2 h, P < 0.05, N = 6-7). Sweet potato (Ipomoea batatas L.) plants were cultivated in vitro in Murashige and Skoog (MS) medium with 20 and 40 g L-1 of sucrose under two different photon flux densities (21 and 60 µmol m-2 s-1). Leaves developed in vitro mostly showed high variable to maximum fluorescence ratio (F V/F M) , indicating good development of photosynthetic apparatus. This ratio decreased during leaf aging, especially in the plants cultivated in vitro on medium with higher sucrose concentration and higher photon flux. The present research aimed to characterize some biochemical responses of Coffea canephora (clones 02 and 153) and C. arabica (Catucaí IPR 102) genotypes subjected to low positive temperatures, helping to elucidate the mechanisms involved in cold tolerance. For that, one year old plants were subjected successively to 1) a temperature decrease (0.5°C a day) from 25/20°C to 13/8°C (acclimation period), 2) a three day chilling cycle (3x13/4°C) and to 3) a recovery period of 14 days (25/20°C). Compacted mouse morulae were frozen at 0.3ºC/min. or 0.5ºC/min. from -6ºC to -24ºC or -32ºC in 10% of glycerol plus different sucrose concentrations with or without 0.1% of honeybee royal jelly. Embryos were thawed in water bath at 22ºC for 20 seconds and cryoprotectant dilution was done in three steps. Effects of culture conditions on the production of inulinase by Kluyveromyces marxianus BABT

The present study was conducted to investigate the influence of initial sucrose concentration, pH and aeration rate on biomass and inulinase production by Kluyveromyces marxianus var. bulgaricus in a stirred batch reactor. Maximum inulinase activity (15.29 UmL-1) was obtained at a sucrose concentration of 10 g L-1, pH 5.0 and aeration rate of 1 vvm. The 20 g L-1 sucrose concentration was suitable for cell growth; however, enzymatic activity at this concentration was inhibited due to catabolic repression. The increase in aeration rate caused a reduction in enzyme activity with no relevant biomass increase.

Effects of preculture with sucrose and aba on cell suspensions water status and its relation with vitrification resistance Rev. Bras. Fisiol. Veg.

Changes in cell water relations during precultures were followed in an attempt to understand the mechanism of cell hardening for cryopreservation by vitrification. Medium containing 0.4 M sucrose (psiw=-1.45 MPa) and containing 5 mg L-1 of ABA (MT psiw=-0.73 MPa and MS psiw=-0.48 MPa) were used to harden cell suspensions of orange and carrot. Preculture in these medium did not cause a significant decrease of cell viability, however, it improved the cell survival to PVS2 and liquid nitrogen expositions.
